Abstract

INDONESIA: Katarak adalah kondisi kekeruhan dari lensa mata atau hilangnya transparansi lensa mata yang dapat menyebabkan gangguan atau penurunan penglihatan. Dunaliella salina merupakan salah satu mikroalga yang memiliki banyak zat bioaktif dan mampu mengakumulasi β-karoten tinggi yang memiliki potensi sebagai antioksidan eksogen yang dapat dijadikan sebagai pencegah stress oksidatif yang tentunya akan mampu mencegah katarak dan dapat menekan reaksi berantai radikal bebas dalam tubuh. Penelitian ini untuk mengetahui efek penambahan D. salina terhadap pembentukan katarak mencit (Mus musculus) yang diinduksi naftalen. K- diberi aquades. K+ diberi naftalen 1 g/Kg BB/hari. K1 diberi Vitamin E 378 IU/Kg BB/hari dan naftalen 1 g/Kg BB/hari. K2 diberi biomassa D. salina 2 g/Kg BB/hari dan naftalen 1 g/Kg BB/hari. Mencit mendapat perlakuan selama 28 hari secara oral. Hasil penelitian menunjukan bahwa mikroalga D. salina mengandung alkaloid, steroid, saponin, asam askorbat dan tannin. Hasil uji DPPH diketahui mikroalga D. salina memiliki Inhibitory Concentration (IC50) sebesar 10,06 µg/mL artinya memiliki antioksidan yang sangat kuat. D. salina mampu menghambat terbentuknya katarak sebanding dengan perlakuan vitamin E yang terlihat pada makroskopis lensa dengan tingkat kekeruhan sebesar 1%. Begitupun hasil gambaran histopatologi menunjukan kerusakan yang kecil pada lensa mata mencit. Kadar protein lensa mata mencit yang mendapat perlakuan mikroalga D. salina lebih tinggi (4,9 x 10-4 µg/mL) dibandingkan perlakuan naftalen (4,7 x 10-4 µg/mL) dan meningkatkan kadar air sebesar 53,81% dibandingkan perlakuan naftalen 46,25%. ENGLISH: Cataract is a condition of opacification of the eyepiece or loss of transparency of the eyepiece that may cause impairment or decreased vision. Dunaliella salina is one of the microalgae that has many bioactive substances and is able to accumulate very many β-carotene which has potential as exogenous antioxidant that can be used as a preventative of oxidative stress which will of course be able to prevent cataract and can suppress free chain reaction of free radicals in the body. This research is to know the effect of addition of D. salina to the formation of naphthalene induced cataract (Mus musculus). K- was given aquades. K+ was given naphthalene 1 g / Kg BW / day. K1 was given Vitamin E 378 IU / Kg BB / day and naphthalene 1 g / Kg BW / day. K2 was given biomass D. salina 2 g / Kg BW / day and naphthalene 1 g / Kg BW / day. Mice received treatment for 28 days orally. The results showed that D. salina microalgae contains alkaloids, steroids, saponins, ascorbic acid and tannins. The result of DPPH test is known that D. salina microalgae having Inhibitory Concentration (IC50) of 10,06 μg / mL means having a very strong antioxidant. D. salina is able to inhibit cataract formation in proportion to vitamin E treatment seen in macroscopic lenses with a turbidity level of 1%. Likewise histopathologic results show little damage to the lens of the mouse eye. Mice lens protein levels obtained by D. Salina microalgae treatment were higher (4.9 x 10-4 μg / mL) than naphthalene treatment (4.7 x 10-4 μg / mL) and increased water content by 53.81% naphthalene treatment of 46.25%.
