Abstract

INDONESIA Anggrek D. taurulinum merupakan anggrek endemik pulau seram yang dapat djadikan sebagai potensi pasar tanaman hias khas Indonesia. Upaya untuk meningkatkan keragaman tersebut, induksi poliploid merupakan solusi yang dapat dilakukan. Pemberian Kolkisin pada tanaman mampu menghambat pembelahan sel. Penelitian ini bertujuan untuk mengetahui pengaruh interaksi konsentrasi dan lama perendaman kolkisin terhadap pertumbuhan tanaman Anggrek D. taurulinum hasil induksi poliploid secara in vitro. Penelitian dilakukan di laboratorium Fakultas Pertanian Universitas Winayamukti Tanjungsari, dari bulan April sampai dengan bulan Juli, menggunakan Rancangan Acak Lengkap (RAL) 2 faktorial dengan 3 kali ulangan. Faktor pertama adalah konsentrasi kolkisin yang terdiri dari 4 taraf : k0 = 0,00 ml. L-1(Kontrol), k1 = 0,25 ml. L-1, k2 = 0,50 ml. L-1, dan k3 = 0,75 ml. L-1. Faktor kedua adalah lama perendaman protocorm menggunakan kolkisin terdiri atas 3 taraf : p1 = 24 jam, p2 = 48 jam, p3 = 72 jam. Parameter pengamatan yang dilakukan meliputi tinggi planlet, jumlah tunas, jumlah daun, warna daun, waktu awal muncul tunas dan uji morfologi stomata. Hasil penelitian menunjukan terjadi interaksi antara konsentrasi dan lama perendaman kolkisin pada parameter tinggi planlet, jumlah tunas dan morfologi stomata. Konsentrasi 0,25 mL L-1 dan lama perendaman kolkisisn selama 72 Jam merupakan perlakuan yang paling baik dalam menginduksi planlet menjadi tanaman poliploid. ENGLISH D. tauralinum orchid is an endemic orchid on Seram island that can be made into potentialy selling plant in the native Indonesian ornamental plants market. In order to increase the diversity, poliploid induction is a solution that can be done. Giving colchicine into the plant can detain the dichotomy of cells within the plant. This Study aimed to determine the effect of interaction of concentration and duration of immersion time on the growth of orchid plants D. taurulinum as a result of polyploid induction using colchicine in in vitro. The Research was conducted in the laboratory Faculty of Agriculture Winaya Mukti University Tanjungsari, from April until July, using factorial completely randomized design with 3 replications consists of 4 level k0 = 0,00 mL.L-1 (Control), k1 = 0,25 mL.L-1, k2 = 0,50 mL.L-1 , and k3 = 0,75 mL.L-1. The second factor was protocorm immersion time using colchicine consisting of 3 levels : p1 = 24 hours, p2 = 48 hours, p3 = 72 hours. Parameters of observation including plant height, number of shoots, number of leaves, color of leaves, budding time appears and analysist morfologi of stomata. The results of this research showed an interaction between concentration and immersion time of colchicine on parameters of plantlet height, number of shoots and stomata morphology. The concentration of 0.25 mL L-1 and immersion time for 72 hours is the best treatment for inducing plantlets into polyploid plants.
