Abstract

Produk kosmetika telah dianggap sebagai barang penting dalam kehidupan sehari-hari. Akibat kebutuhan pasar yang meningkat akan produk kosmetika, industri kosmetika terus berkembang. Atas dasar permintaan pasar yang tinggi ini menyebabkan banyaknya masuk produk kosmetika dari negara lain masuk ke Indonesia melalui kegiatan impor. Produk kosmetika yang paling sering dijumpai adalah lipstick, lipbalm, dan krim wajah ketiga produk tersebut beredar di berbagai wilayah Indonesia dan beberapa diantaranya tidak memiliki logo halal. Bagi umat muslim sangat penting untuk mengetahui komposisi, bahan mentah dan proses dari sebuah produk yang digunakan. Salah satu komposisi bahan yang digunakan dari pembuatan lipstick, lipbalm, dan krim wajah adalah gliserin, gelatin, dan kolagen dimana ketiganya dapat berasal dari turunan babi seperti lemak atau plasentanya. Untuk mengidentifikasi kandungan DNA babi pada lipstick, lipbalm dan krim wajah dapat menggunakan metode Polymerase Chain Reaction (PCR). Tujuan dari penelitian ini adalah mendeteksi kandungan babi pada lipstick, lipbalm, dan krim wajah tanpa logo halal yang berasal dari luar Indonesia. Dua sampel lipstick, dua sampel lipbalm, dan dua sampel krim wajah yang digunakan dipilih berdasarkan purposive sampling. Ekstraksi DNA pada sampel dilakukan dengan menggunakan Dneasy Mericon Food Kit. Proses PCR menggunakan primer sitokrom b dengan suhu annealing yang telah dioptimasi yaitu pada 62,3°C. Hasil yang didapatkan divisualisasikan dengan elektroforesis gel agarosa. Elektroforegram menunjukkan adanya pita dengan ukuran sekitar 131 bp pada keempat sampel, sehingga lima dari enam sampel dideteksi mengandung babi. Cosmetic products have been considered as important items in everyday life. Due to the increasing market demand for cosmetic products, the cosmetics industry continues to grow. Based on this high market demand, many cosmetic products from other countries enter Indonesia through import activities. The most frequently encountered cosmetic products are lipstick, lip balm, and face cream, these three products are circulating in various parts of Indonesia and some of them do not have a halal logo. For Muslims it is very important to know the composition, raw materials and processes of a product used. One of the ingredients used in the manufacture of lipsticks, lip balms, and face creams is glycerin, gelatin, and collagen, all of which can be derived from pork derivatives such as fat or placenta. To identify the content of pig DNA in lipstick, lip balm and face cream, the Polymerase Chain Reaction (PCR) method can be used. The purpose of this study was to detect pork content in lipstick, lipbalm, and face cream without a halal logo originating from outside Indonesia. Two samples of lipstick, two samples of lip balm, and two samples of face cream used were selected based on purposive sampling. DNA extraction in samples was carried out using the Dneasy Mericon Food Kit. The PCR process uses cytochrome b primers with an annealing temperature that has been optimized at 62.3°C. The results obtained were visualized by agarose gel electrophoresis. The electrophorogram showed a band with a size of about 131 bp in the four samples, so that five of the six samples were detected to contain pork.
