Abstract

Indonesia : Diabetes mellitus merupakan suatu penyakit metabolik yang ditandai oleh tingginya kadar glukosa didalam darah akibat dari defesiensi atau kerusakan dalam sekresi insulin. Berdasarkan penyebabnya diabetes mellitus terbagi menjadi beberapa jenis yaitu diabetes mellitus tipe I, diabetes mellitus tipe II, dan diabetes gestasional. Hingga saat ini kasus diabetes mellitus tipe 2 merupakan kasus terbesar di Indonesia karena disebabkan oleh berubahnya gaya hidup manusia yang terus meningkat seiring dengan meningkatnya tingkat kemakmuran. Pengobatan diabetes mellitus tipe 2 telah banyak dilakukan yaitu dengan menggunakan obat oral antidiabetik dan injeksi insulin akan tetapi memiliki efek samping yang cukup mengganggu. Salah satu mekanisme yang paling efektif dalam pengobatan diabetes mellitus tipe 2 yaitu dengan menghambat enzim α-Glukosidase pada saluran pencernaan untuk memperlambat pembebasan d-glukosa. Beberapa senyawa golongan flavonoid seperti senyawa apigenin, kuersetin, dan xantone dipercaya bermanfaat dalam pengobatan diabetes mellitus tipe 2. Oleh karena itu diperlukan pengujian dalam merancang sebuah obat melalui pendekatan metode In Silico sebagai komplemen dari metode In Vitro dan In Vivo. Tujuan penelitian ini yaitu mengetahui nilai perbandingan binding affinity senyawa uji Apigenin, Kuersetin, dan Xantone dalam berikatan pada reseptor α-glukosidase dan mengetahui senyawa yang paling berpotensi sebagai obat antidiabetes mellitus tipe 2. Penelitian ini dilakukan di Laboratorium Pengolahan Data Proses Jurusan Biologi Fakultas Sains dan Teknologi, Universitas Islam Negeri Sunan Gunung Djati Bandung dan dilaksanakan pada bulan Maret hingga Juni 2020. Hasil penelitian ini didapatkan perbandingan nilai Binding Affinity dan aktivitas kimia dari ketiga senyawa uji yaitu senyawa apigenin sebesar -8.5 kcal/mol dengan aktivitas kimia yang stabil, senyawa kuersetin sebesar -7.4 kcal/mol dengan aktivitas kimia yang kurang stabil, dan senyawa xantone sebesar -7.1 kcal/mol dengan aktivitas kimia yang kurang stabil. Berdasarkan hal tersebut, senyawa apigenin paling berpotensi sebagai senyawa antidiabetes mellitus tipe II dengan target reseptor α-glukosidase sebab memiliki nilai binding affinity terkecil dan mempunyai aktivitas ikatan-ikatan kimia yang lebih stabil. English : Diabetes mellitus is a metabolic disease characterized by high levels of blood glucose caused by deficiency of in insulin secretion. Based on the case, diabetes mellitus is divided into several types, there are diabetes mellitus type I, diabetes mellitus type II, and gestational diabetes. Nowadays, diabetes mellitus type 2 was the biggest case in Indonesia it is caused by changes of human lifestyles as long as increasing level of prosperity. The treatment for diabetes mellitus type 2 has been done by using oral antidiabetic drugs and insulin injection but it has a silent side effects. One of the most effective ways in the treatment of type 2 diabetes mellitus is that it inhibits α-Glucosidase enzyme in digestive tract to remove d-glucose impurities.. Several flavonoid class compounds such as apigenin, quercetin, and xanthon compounds are believed to be useful in the treatment of type 2 diabetes mellitus.Therefore, testing is needed in designing a drug through In Silico method approach as a complement to the In Vitro and In Vivo methods. The purpose of this study is to determine the comparative value of the biding affinity of the test compounds Apigenin, Quercetin, and Xanthon in binding to the α-glucosidase receptor and to determine which compounds which have the most potential as type 2 antidiabetic mellitus drugs. This research was conducted at the Process Data Processing Laboratory, Department of Biology, Faculty of Science and Technology, UIN Sunan Gunung Djati Bandung and implemented from March to June 2020. The results of this study obtained a comparison of the value of binding affinity and chemical activity of the three test compounds, namely apigenin compounds of -8.5 kcal / mol with stable chemical activity, quercetin compounds of -7.4 kcal / mol with unstable chemical activity, and xantone compounds of - 7.1 kcal / mol with less stable chemical activity. Based on this, the apigenin compound has the most potential as an antidiabetic mellitus type II compound targeting the α-glucosidase receptor because it has the smallest binding affinity value and has more stable chemical bonding activity.
