Abstract

INDONESIA Fosfat dikarst masih dalam bentuk sedimen yang berupa batuan beku tersedia sekitar 0,12%. Pemanfaatan fosfat dibidang pertanian memerlukan bantuan bakteri pelarut fosfat (BPF) setelah itu bisa digunakan sebagai nutrisi oleh tumbuhan. Identifikasi molekuler mengunakan 16s rRNA, molekul 16s rRNA memiliki beberapa daerah yang memiliki urutan basa yang relatif konservatif sekitar 1500-1600 basa nukleotida. Tujuan penelitian ini untuk mengetahui spesies isolat bakteri (K2.BR.5) yang diisolasi dari rhizosfer tanaman Imperata cylindrica kawasan karst Citatah Padalarang Kabupaten Bandung. Penelitian meliputi. Ekstraksi DNA, Amplifikasi DNA dengan PCR, uji kualitatif DNA dengan elektroforesis dan Sekuensing. Hasil penelitian diperoleh konsetrasi sampel 115.1 ng/µl dengan kemurniaan DNA 0,82 pada panjang gelombang A260/280, kemudian hasil PCR pita DNA diuji kualitatif dengan elektroforesis dengan panjang fragmen 1500 bp, gen 16s rRNA hasil pada pohon filogenik bakteri Isolat K2.BR.5 memiliki kemiripan atau kekerabatan dengan spesies Aneurinibacillus migulanus Isolate Am25 dengan nilai bootstrap 99 %. Berdasarkan jarak genetik antara Isolat K2.BR.5 dengan Aneurinibacillus migulanus Isolate Am25 memiliki nilai jarak sebesar 0.712 %. Isolat K2.BR.5 merupakan Spesies Sp. dengan panjang fragmen 1500 bp pada 16s RNA. ENGLISH Phosphate in karst is still in form of sediments that formed by igneous rock provided around 0.12%. Utilization of phosphate in agriculture need phosphate solvent bacteria (BPF) then could be used as nutrients by plants. Molecular identification using 16S rRNA, 16S rRNA molecules have several areas that have a relatively conservative base sequence of nucleotide bases around 1500-1600. The purpose of this study to determine the species of bacteria (K2.BR.5) were isolated from the rhizosphere of plants Imperata cylindrica in karst region Citatah Padalarang Bandung regency. The study covers DNA extraction, DNA amplification by PCR, qualitative test DNA by electrophoresis and sequencing. The results are concentration of sample is 115.1 ng/mL with DNA purity 0.82 at wavelength A260/280. Then the DNA from PCR tested qualitatively by electrophoresis with a length of 1500 bp fragment. Based on filogenik tree gene 16S rRNA in isolates has a likeness with the species Aneurinibacillus migulanus Isolate Am25 with bootstrap value 99%. While the gene distance between them is 0.712%. Isolates K2.BR.5 is Species Sp. with a fragment of 1500 bp in length 16s RNA.
